Glial cell line-derived neurotrophic factor (GDNF) is a disulfide-linked homodimeric glycoprotein that belongs to the TGF-β superfamily. It has been shown to promote the survival of different subpopulations of neurons in both the central and peripheral nervous systems at various stages of development. Human GDNF cDNA encodes a 211 amino acid precursor, which is processed to produce the dimeric protein. Mature human GDNF is expected to contain two subunits, each with 134 amino acid residues. Cells known to express GDNF include supporting cells, type I astrocytes, Schwann cells, neurons, pinealocytes, and skeletal muscle cells. Mutations in this gene may be associated with congenital megacolon.

Mature human IGF-1 consisting of 70 amino acids has 94% identity with mouse IGF-1 and exhibits cross-species activity. IGF-1 binds IGF-IR, IGF-IIR, and the insulin receptor and plays a key role in cell cycle progression, cell proliferation and tumor progression. IGF-1 expression is regulated by growth hormone.
R3 IGF-1 is an 83 amino acid analog of IGF-1 comprising the complete human IGF-1 sequence with the substitution of an Arg (R) for the Glu(E) at position three, hence R3, and a 13 amino acid extension peptide at the N terminus. R3 IGF-1 has been produced with the purpose of increasing biological activity. R3 IGF-1 is significantly more potent than human IGF-1 in vitro.

Noggin is a secreted homodimeric glycoprotein that is an antagonist of bone morphogenetic proteins (BMPs), Noggin antagonizes the biological activity of BMP by blocking the binding of BMP to type I and II receptors. Noggin shows a high affinity to BMP4 but a low affinity to BMP7. During embryogenesis, Noggin antagonizes specific BMPs at defined times, for example, during neural tube, somite and cardiomyocyte growth and patterning. During bone development, Noggin prevents chondrocytes from proliferating so as to allow normal joint formation. During culture of human embryonic stem cells (hESC) or neural stem cells under certain conditions, addition of Noggin to antagonize BMP activity may allow stem cells to proliferate while maintaining their undifferentiated state, or alternatively, to differentiate into dopaminergic neurons. Noggin is expressed in defined areas of the adult central nervous system and peripheral tissues such as lung, skeletal muscle and skin.

Regardless of gender, RSPO1 is required for early development of Gonad. This phenomenon was discovered only 11 days after fertilization in mice.
