DNase I (Deoxyribonuclease I) was initially isolated from bovine pancreas. It can randomly degrade single stranded or double stranded DNA with equal efficiency, generating oligonucleotides with 5’-P. The bioactivity of DNase I is highly dependent on Ca2+, Mg2+ or Mn2+. In the presence of Mg2+, DNase randomly cuts dsDNA; in the presence of Mg2+, DNase cleaves both strands of dsDNA at approximately the same site, resulting in a blunt end or a sticky end with 1 or 2 protruding nucleotides.  DNase I is GMP Grade produced in Pichia pastoris. Various enzymes are used in the process of in vitro transcription and modification of mRNA. As a protein substance, enzymes can be identified by Dot Immunobinding Assay (DIBA, 2020 edition, Part IV, General Principles 3402). This kit uses polyvinylidene fluoride (PVDF) as the solid phase, carries out antigen-antibody reaction by DIBA, and carries out the identification test of various raw materials. The kit contains six antibodies, anti-T7 RNA Polymerase antibody, anti-Pyrophosphatase Inorganic (Yeast) antibody, anti-RNase Inhibitor antibody, anti-DNase I antibody, anti-Vaccinia Capping Enzyme antibody and anti-mRNA Cap 2´ -O-Methyltransferase antibody. The characteristics of RNase Inhibitor are similar to those derived from pig liver and human placenta. The inhibition is reversible, as the complex can be dissociated by urea and sulfhydryl reagents in which RNase refolds and the inhibitor is inactivated. It can be directly added to the reaction containing RNA. Different from other competitive inhibitors (e.g., nucleic acids, inorganic phosphoric acids), this product is a protein, which can be easily removed from the reaction by phenol extraction. The vaccinia capping enzyme together with substrates, is capable to add the 7-methylguanosine capping structure (m7Gppp, Cap0) to the 5´ end of the RNA. In eukaryotes, this structure is closely related to the stability, transport, and translation of mRNA. Capping RNA with an enzymatic reaction is a simple and effective method that can significantly improve the stability and translation capabilities of RNA used for in vitro transcription, transfection, and micro-injection. The vaccinia virus capping enzyme consists of two subunits (D1 and D12) (Figure 1a), and possesses RNA triphosphatase, guanosine transferase, and guanine methyltransferase activities, and they are all necessary for adding a complete Cap0 structure m7Gppp5´N to mRNA. This product can be applied to capping reaction of RNA generated by the T7 RNA Polymerase. The capping reaction is completed within an hour and the efficiency is close to 100% with correct direction.
