Transforming growth factor (TGF-beta 1 or TGF-β) is a multifunctional cytokine with an important regulatory effect on cell growth, differentiation, and immune function. It can both activate or inhibit the immune system. The tumor inhibitory effect of TGF-beta signaling derives from its ability to induce the expression of multiple genes. These genes are involved in inhibiting cell proliferation, inducing apoptosis, activating autophagy, inhibiting growth factor signaling through stromal fibroblasts, inhibiting inflammation, and inhibiting angiogenesis. CD40L, also known as CD154, is a plasma membrane glycoprotein and differentiation antigen expressed on the surface of T cells. It stimulates the proliferation of B cells to secrete various immunoglobulins in the presence of cytokines. CD40L can induce the cytokines secretion of peripheral blood mononuclear cells, so produce anti-tumor activity. It also has a co-stimulatory effect on the proliferation of activated T cells, accompanied by the production of IFN-gamma, TNF-alpha, and IL-2. Vitronectin is a large glycoprotein found in blood and the extracellular matrix (ECM). The gene for Vitronectin encodes a 19 amino acid (aa) signal peptide and a 459 aa protein. The apparent molecular weight of human Vitronectin is 75 kDa, with ~30% of its molecular mass being attributed to glycosylation at 3 different sites. Vitronectin is involved in a number of biological activities including cell adhesion, cell spreading and migration, cell proliferation, extracellular anchoring, fibrinolysis, hemostasis, and complement mediated immune defense. It is worth noting that in the culture of pluripotent stem cells, due to the clear chemical composition of the protein, Recombinant Human Vitronectin helps to reduce the instability, compared with human plasma-derived Vitronectin and basement membrane extracts (BMEs).
