Ribonuclease R (RNase R), from the RNR superfamily of E.coli, is a Mg²⁺-dependent 3'-5' exonuclease. RNA can be cleaved into dinucleotide and trinucleotide gradually from the 3'-5' direction by RNase R. RNase R can digest most of the linear RNA. However, it is difficult to digest circular RNA, lariat RNA and short double-stranded RNA molecules with less than 7 nucleotides of 3' end protrusion. RNase R is commonly used in gene expression and alternative splicing studies and digests linear RNA to enrich circular RNA or lariat RNA. RNase Inhibitor inhibits RNase A, B and C with high efficiency by forming a 1:1 complex with them. The characteristics of RNase Inhibitor are similar to those derived from pig liver and human placenta. The inhibition is reversible, as the complex can be dissociated by urea and sulfhydryl reagents in which RNase refolds and the inhibitor is inactivated. It can be directly added to the reaction containing RNA. Different from other competitive inhibitors (e.g., nucleic acids, inorganic phosphoric acids), this product is a protein, which can be easily removed from the reaction by phenol extraction. However, the reverse transcriptase RNase H could not be inhibited. Recombinant RNase Inhibitor is GMP Grade produced in E. coli. S-(5’-Adenosyl)-L-methionine (SAM, AdoMet) with an activating methyl group is the methyl donor for vaccinia capping enzyme and mRNA Cap 2’-O-Methyltransferase catalyzed methylation, the storage buffer formula is: 0.005M Sulfuric Acid, 10% ETOH.
